Intravital microscopy can visualize various in vivo cellular-level dynamics such as cell trafficking, cell-to-cell or cell-to-microenvironment interactions in a living animal. Intravital imaging of pathophysiological cellular dynamics in natural in vivo microenvironment in various preclinical animal model can provide an unprecedented insight in the dynamic processes of human diseases beyond conventional histological observation of ex vivo sample or in vitro culture sample.

In this talk, a real-time laser-scanning intravital confocal/two-photon microscopy system will be introduced. The imaging system has been extensively optimized for in vivo cellular-level imaging of internal organs in live animal model for various human diseases, which can acquire a real-time multi-color fluorescence microscopic image in sub-micron resolution. Intravital microscopic imaging of various organs including skin, liver, spleen, pancreas, kidney, small intestine, colon, retina, lung, heart, lymph node, and bone marrow will be briefly introduced.

ACCELA Educational webinar series: NanoView - Purification-free characterization of extracellular vesicles and viruses

NanoView Biosciences has developed a unique technology for purification-free analysis of extracellular vesicles and viruses. Exosomes are affinity captured onto a protein microarray (ExoView chip) via common or custom exosomal markers.

Once bound to the surface of the ExoView chip each individual exosome can be sized, counted and characterized in terms of protein expression. Fluorescent antibodies can be added (in 3 channels so that up to 4 proteins can be measured on individual exosomes with single-molecule sensitivities meaning that even the smallest of exosomes and can be detected and characterized. Exosomal cargo can also be detected through the application of cargo protocols to detect proteins not expressed on the surface of exosomes.

In this webinar, Professor Bijoy Kundu will describe the application of PET imaging for understanding the early metabolic changes that arise in animal models of hypertension-induced LVH and heart failure. Prof. Kundu and team have developed dynamic/kinetic imaging techniques and obtaining time-resolved images from PET scanners to assess the metabolic changes that occur in the heart when it is subjected to pressure overload. Clinicians might be interested in using PET to detect metabolic changes in patients with early stages of heart failure, with a view to prevent the structural changes that are seen later with MRI.

Abstract: Current assessment of human immunotherapy agents prior to clinical trials is by use of assays that allow only cancer and T-cell interactions (in vitro cytotoxicity assays with labeled cancer cells and in vivo anti-tumor efficacy studies in immunodeficient mice). By use of the xCELLigence impedance assay, we have optimized an ex vivo culture system with patient T cells and malignant pleural effusions. This system measures dynamic, long-term interactions of CAR T cells, cancer and immune cells and provides sequential CAR T-cell efficacy and phenotypic changes.

During the years of research it has been proven that oxygen and pressure have distinct physilogical effects and both play important but idependent roles in the cellular microenvironment. Under hypoxia and pressurized cultuing conditions, changes are seen in cell morphology, gene and protein expression. Progress in plant genome research is vital to understanding natural evolution and feeding the world. The size and highly repetitive nature of plant genomes make building quality reference genomes a challenge for short-read sequencing technologies. The combination of Bionano maps with sequencing data improves assembly accuracy and quality while reducing the need for deep sequencing coverage. This improved contiguity significantly lowers costs associated with assembling reference genomes.
